GWCCG Flow Facility Instrument Profile 

GWCCG Flow Facility harbours state-of-the-art simple and high-parameter flow cytometers for cell analysis and sorting and is always improving to help researchers achieve their goals.


GWCCG Flow Sorters

The BD FACSAria™ III Cell Sorters is a high-speed fixed-alignment benchtop cell sorter that has 5 excitation lasers and can detect up to 19 parameters.

Excitation lasers:

  • Violet 405nm
  • Blue 488nm
  • Yellow/Green 561nm
  • Red 633nm
  • UV 355nm

The Aria III Hooded instrument at GWCCG is located within a biological safety cabinet to enable the sorting of human patient samples.

Runs on Diva 8 software and equipped with 2-way, 4-way, 6-well, 12-well, 96-well and 384-well plate sorts.

GWCCG Flow Analyzers

The BD FACSymphony is a high throughput analysing cytometer that has eight excitation lasers:

  • UV 355 nm
  • Violet 402 nm
  • Blue 488 nm
  • Blue/Violet 442 nm
  • Yellow/green 561nm
  • Red (628 nm)/Orange (592 nm)
  • NIR 785 nm
  • The FACSymphony is our newest analysing cytometer with the capacity to analyse 50 colours using the most cutting edge technology BD has to offer. Compared to alternative cell analysis system , the BD FACSymphony instrument offers high-speed analysis with detection rates of up to 40000 events per second.
  • This advanced platform features an ultra-quiet electronics system adapted that improves detection sensitivity to enable the identification and analysis of rare cell types and events.

The BD LSR Fortessa is a 4 Laser Cell Analyser with ability to detect up to 18 parameters (16 fluorochromes + Side scatter+ forward scatter) to run a comprehensive multi-colour flow cytometry assay. 

Excitation lasers:

  • Blue 488 nm
  • Yellow/Green 561 nm
  • Red 633 nm
  • UV 355nm

 

The BD LSR SORP is a high end cell analysing cytometer that has five excitation lasers (discontinued):

  • UV355nm
  • Violet 405nm
  • Blue 488nm
  • Yellow/green 561nm
  • Red 640nm
  • The SORP is our most popular cell analysis instrument as it has a solid solid state UV laser that can detect dyes with low excitation wavelengths (e.g. Hoescht, DAPI and Indo-1). The addition of the UV lasers enables the user to expand their fluorochrome panel to up to 19 (17 fluorochromes + Forward Scatter + Side Scatter).
  • The SORP also has a green laser (532nm) that can provide even more flexibility with designing a robust fluorochrome panel with minimal spectral overlap. Please contact us if you require the green laser in your experiments.
  • Runs on FACS Diva 8.
  • The instrument is now discontinued from BD.

 

The BD FACSCantoTM II system is a compact flow cytometer that allows quick sample acquisition, fully automated fluidics start up that is designed to streamline work, save time, and improve performance.

The FACS Canto II has 3 excitation lasers:

  • Violet 405nm
  • Blue 488nm
  • Red 633nm

 

  • The Canto II is capable of detecting up to 10 parameters (8 fluorochromes + Forward Scatter + Side Scatter).
  • The Canto II runs on FACS Diva 8.

The Beckman Coulter Cytoflex is a compact benchtop flow cytometer that is equipped with 3 lasers and can detect up to 13 parameters (including forward/side scatter). 

Excitation lasers:

  • Violet 405nm
  • Blue 488nm
  • Red 640nm

 

  • This instrument features a fully automated start up procedure and innovative fluidics system that minimises waste and sheath required for acquisition. The Cytoflex tube loader acquires samples from FACS tubes.
  • The Cytoflex plate reader is equipped with the CytExpert Software program mentioned on the tube analyser page which has features such as auto-thresholding and a compensation library for quick access to previously stored compensation matrices.
  • The Cytoflex software 'CytExpert' has numerous useful features:

    • A compensation library to store a repository of compensation control panels in a libraryfor easy access to the correct compensation matrix with new gain settings.
    • Auto thresholding to easily find target populations. Manual thresholding can also be selected.
    • Axis Pan which is an ideal tool for visualizing data across a high dynamic range axis. You can easily pan across the axis range to search if there is important data out of range.
